Experimental validations highlighted TaDOF5.6 ( TraesCS6A02G274000) and TaDOF3.4 ( TraesCS2B02G592600) as potential enhancers of transformation efficiency in different wheat varieties. Further comparisons between wheat and Arabidopsis revealed distinct patterns of DNA binding with one finger (DOF) TFs in the two species. The built-up TRN driving wheat regeneration was found to be dominated by 446 key transcription factors (TFs). Our results demonstrate that the sequential expression of genes mediating cell fate transition during regeneration is induced by auxin, in coordination with changes in chromatin accessibility, H3K27me3 and H3K4me3 status. RNA-seq, ATAC-seq and CUT&Tag techniques were utilized to profile the transcriptional and chromatin dynamics during early regeneration from the scutellum of immature embryos in the wheat variety Fielder. Here we employed a multi-omic analysis strategy to uncover the transcriptional regulatory network (TRN) responsible for wheat regeneration. Genetic transformation is important for gene functional study and crop improvement.

What's more taunting deals with the fact that a few bonding scenes between certain characters are lived out so damn short. The joy and smiles from the faces of our main characters are wiped forever so don't expect any happy scenes in this episodes. Happiness though? Honestly, there's none of that anymore. Corpse Party paints such dark themes as the way it conveys itself from the premise. Rather than a ghost story, this might be the final chapter for our characters' lives. The deaths and tortures are real and there's no turning back. It is from there on that we immediately take notice just the grievous situation our characters are in. In particular, the first episode sets up a graphic premise for a particular girl named Naomi after she witnesses a gruesome death of her friend. The taunting scenes of blood and body parts carves a haunting memory into the characters. Because the characters are trapped in this new environment of darkness, they struggle to survive both physically and mentally. There is a sense of hopelessness in Corpse Party. This series is about watching kids getting murdered. I'll just throw a warning sign right now that if you can't take the first episode (or rather the first 20 minutes), then you might not have enough stomach to make it. However, they soon become haunted by the ghosts themselves after the earth shaken and sucks them deep down into the abyss.of the once existent school known as Heavenly Host Elementary School.Īs this is a horror theme series, expect dark themes such as blood, gore, and death. based off the game and chronicles the events in which a group of young students tells ghost stories at a school known Kisaragi Academy. (that is if and only if you escape alive)Ĭorpse Party: Tortured Souls - Bougyakusareta Tamashii no Jukyou (or simply known as Corpse Party: Tortured Souls) is a series of four OVA episodes. For once you're trapped, Corpse Party will take you to a place where your life will never ever be the same again. When you realize that death is coming before you, screams are shouted out but you won't be heard. Can't even shout, can't even cry, the dangers are coming by. Watching Corpse Party is literally like watching a horror movie. I do genuinely believe that there aren't many series that pulled off this sort of thing nearly as well, and definitely not in the same time frame as Corpse Party. it was a very genuinely well made horror series that kind of goes away from the norm. Half of the things that happened with this character I didn't see coming and I just love her voice actress.īut for my overall enjoyment. But the character of Sachiko was pretty original. Though, to be fair, it is harder to create a cast of characters within this genre of horror that is original while also keeping it in a more modern time line, you could of done better than taking the generic cast of highschool students (2 possible lesbians, 1 guy with glasses, 1 hopelessly brave guy, the little sister, the occult freak, and the guy with no defining features what so ever, otherwise know as the boring guy). The place where this series is a bit lacking is the unoriginal cast of characters. You can really see the inspiration from Akira Yamaoka within the soundtrack and I thought that it was genuinely a good choice to go that way for a horror series. The opening theme was pretty good and most of the background sound, or even the lack of sound at points was a good choice for the series. I did notice some similarities between this and the Team Silent Silent Hill game's art, what with blood coming down the walls and bodies everywhere, and that was generally a good choice, just to make the series that little bit more disturbing. The art in this series did such a good job at making the art disturbing and grotesque that I almost vomited a couple times throughout the series. I also want to bring in the concept of how good the disturbing art was. This is amazing for that small length of time. But what I really want to emphasis on is the fact that series fit everything into 4, half-hour long episodes. It had a story and a conclusion and we were not left with a Where as this is still not my favorite horror anime, I still think that it did an amazing job with what little time it had. This is one of those series that makes you cringe but you still love it because it takes you on a well written story. Not only is this the most disturbing animated series I have ever watched, it is the most disturbing fiction work I have had the pleasure of experiencing. Well this was probably the most disturbing thing I have ever seen in my life. It's a choice that none of these characters have had - have been able to avail themselves of. And both sides are willing to destroy people to keep going, which is why that moment when Paige steps off the train is so wrenching because it's something she chooses. Both sides on this struggle are playing with human connections, exploiting human connections like love and lust and friendship, as they - as we heard in that clip. And again and again, we've seen this system chew up characters that we love, that we don't know. You were expecting this finale to be explosive. But, man, everything you said was exactly correct. GLEN WELDON, BYLINE: Aisha, this was my first pick. So that's my pick - "The Americans." And after rewatching this episode again, I am so excited to go back and rewatch it all from the beginning because, oh, my goodness, one of the greatest shows. So when you can put it in your probably top five of that show itself, to me, that's what makes it such a great series finale. It's not just a great ending, but it's also probably one of the best episodes of the entire series. It's, like, maybe a little more on the nose than "The Americans" usually is. HARRIS: And it's just like - it is so dramatic. And as soon as Elizabeth notices it, at that same exact moment, you hear Bono go, (vocalizing). She's supposed to be going with them to leave the U.S. There's a moment where we have "With Or Without You" playing by U2, and Paige gets off the train. And Elizabeth lets off this like, gasp-slash-guffaw that just - there's just so much inside that little noise that she makes and shows how, unlike Philip, she has always been sort of the more detached and cool personality who was able to separate her morals from what her job was, which is to kill a lot of people in service of the KGB. And it ends with, like, this, planting of a seed that Stan's wife, Renee, might be one of them.Īnd there's just all these other moments, like whether it's Philip saying, we need to leave Henry, our other son, behind because he knows nothing about any of this, and he has a chance to live a normal life. And the way that tension is played is just so absolutely perfect - like, one of the greatest scenes of all time. And then the rest of the scene is Philip and Elizabeth kind of alternating between BS'ing Stan and not being truthful about what they were doing. And after six seasons, you know that he feels that way. HARRIS: First of all, you can just hear the way he says, you made my life a joke. For all these years, my life was the joke, not yours. MATTHEW RHYS: (As Philip Jennings) You were my only friend in my whole life. NOAH EMMERICH: (As Stan Beeman) You made my life a joke. And this scene is just heartbreaking, and I want to just play a little brief moment here where Stan just kind of breaks down. They become really good friends, but Stan has finally figured it out, and he confronts them at gunpoint. And there is an 11-minute sequence where they are confronted by Stan Beeman, who was an FBI agent who lived right next door to them. And this was a show that always took its time. What I love about the finale is that it has everything that we've loved about the rest of the series. You know, they have that - on the surface - perfect, idyllic suburban life in the '80s, but they are, of course, undercover spies. Matthew Rhys and Keri Russell play Philip and Elizabeth Jennings. So here are the co-hosts, Stephen Thompson, Linda Holmes and Aisha Harris sharing their favorite finales.ĪISHA HARRIS, BYLINE: So I came down to a couple picks, and I finally landed upon a show that I've only watched from beginning to end once but has stuck with me even a few years later and is something I will definitely go back and rewatch at some point, which is "The Americans," the very slow-burn FX series about a couple of KGB agents who are living undercover in Virginia. So what are the best ever TV finales? And what makes a finale great anyways? Well, these are the kinds of questions the crew at NPR's Pop Culture Happy Hour podcast like to tackle. But sometimes a show will get it just right. You have to wrap up the story, leave the characters somewhere satisfying, give the fans what they want, you know, up to a point. And, you know, ending a TV series is not easy. Maisel" and "Succession" are all wrapping up this month. The TV shows "Barry," "The Marvelous Mrs. I used to know every Brooks & Dunn song backwards and forwards, but nowadays I can't really tell a Brad Paisley from a Dierks Bentley. Video for Rascal Flatts - "Summer Nights" We're gonna be looking at some country music today. That's right, put on your cowboy boots and crack open an ice cold beer. Can’t wait to get that out there.Todd plays "If I Die Young" on his piano in a very upbeat manner. Will you be releasing a video with this song? I’m a mixed bag when it comes to music haha. But also the Beatles were huge for me in my early teens when I was learning to play guitar and pop rock like Blink-182 and Oasis where my “go to’s” in High School. George Strait, Alan Jackson my biggest influences. I grew up listening to a lot of country with my parents. Has a little bit of an edge to it which is definitely me. For sure it’s different from the pop country that’s all over the radio. I got a lot of positive feedback for this song and a few others. During quarantine, I played a lot of originals on my live streams from the house. It is expensive record but I knew this song needed to be released. I’ve written a lot over the years but never pulled the trigger on investing my own money to track professionally. What sets this song apart from your previous work? What sets this song apart from what is being played on the radio today? Like I said, it wrote itself and the process was super quick. Were there any challenges while writing it? I just wanted to get in the truck and drive! To just escape from the crappy reality of being stuck at home. Lyrically, what perspective is the song told from?įrom my own point of view. Stay tuned! Great song about a girl who’s heartbroken and not looking for any kind of romance. I am going to release a couple more singles this year. I do plan to release a full EP at some point. Is it going to be a part of a larger collection, such as EP or album? I finished the tune within about an hour. I pictured hopping in the truck, driving to the beach, and it just started to flow right out. I sat in bed and even without a voice I decided to try and write a tune. I had just finished a live stream show from my house and I had actually lost my voice and had to end the show early. What came first, the music, the words, the idea? We drove that truck around Tennessee a lot during the shutdown and one night the title/hook line came to mind put it all in the “rearview.” A positive song about getting away from our everyday cares and worries. My inspiration came from my 1972 International Scout II. It really was a miserable time being stuck inside and nothing to do. I wrote the song in April of 2020 right in the thick of the Covid quarantine/shutdown.
